Fabrication of peg-well microarrays on a track-etched membrane forsingle cell selection

    Abstract

    Isolation and expansion of a single productive cell from a large population of non-productive cells is widely used in biotechnology for many different purposes i.e. in monoclonal antibody (Ab) production. Conventional methods need the formation of large colonies, which require weeks to grow. Here, we propose a new microfluidic system for the identification of productive clones using a few cells only, that can be obtained within a few days. A polyethylene glycol (PEG) microwell array with a track-etched membrane as the bottom is used atop a PDMS substrate coated with a capture Ab. Proteins released by cells diffuse through the track-etched membrane, bind to the surface beneath the cell, and may subsequently be localized by incubating the PDMS substrate with a fluorescently tagged detection Ab; the localization of a bright spot corresponds to a productive cell line which can then be retrieved.
